DNA2次元電気泳動自動化システムの開発

 2次元電気泳動法(RLGS:Restriction Landmark Genomic Scanning)は,ヒトのようにゲノムサイズの大きいものにも有用で,がんや変異マウスの遺伝子解析などに応用されている。しかしながら,煩雑でハードな作業であり,実験者は高度なスキルが要求される。また,RIを使用するため,身体が危険にさらされる。そこで,このような問題を解決するために,2次元電気泳動自動化装置と電気泳動パターン読み取り装置の開発を行った。前者は,電気泳動の全工程を自動的に行う装置である。後者の装置は,電気泳動パターン読み取り装置であり,蛍光標識されたDNA断片の2次元電気泳動パターンを検出するものである。RI標識を蛍光標識に代替することにより,安全性と再現性が大幅に向上した。

Development of the Automated DNA Two-dimensional Electrophoresis System and of the Electrophoresis Image Scanner

The RLGS (restriction landmark genomic scanning) method is useful for large genomes such as those of humans, and is used in the analysis of cancer genes, mutant mouse DNA, and so on. However, it involves complicated, difficult operations, and requires high-level skills on the part of technicians. In addition, scientists are exposed to dangerous radiation because the original RLGS method makes use of radioisotopes (RI). In order to solve these problems, we have developed an automated two-dimensional electrophoresis system and an electrophoresis image scanner that does not require the use of radioisotopes. The system conducts all of the procedures for electrophoresis automatically. The scanner detects two-dimensional electrophoresis images of fluorescent-labeled fragments of DNA. Fluorescent labeling has improved the safety and reproducibility of RLGS imaging.
